چکیده

Abstract Study question How does non-invasive preimplantation genetic testing for aneuploidy (niPGT-A) perform to predict sustained implantation or live birth (SI/LB)? Summary answer Though not comparable conventional PGT-A, niPGT-A can still be considered an appealing tool clinical embryo prioritizing given its high negative predictive value (NPV). What is known already To omit the need invasive trophectoderm biopsy, which screens embryonic ploidy using cell free DNA released into spent culture medium (SEM) blastocoel fluid (BF) was developed. Although feasibility of has been shown and healthy births have reported from embryos screened by niPGT-A, transfer outcome established yet evidence lacking as whether selection associated with improved pregnancy outcomes. The answers these are vital importance justify care. design, size, duration This a single center, blinded, non-selection study consisting frozen cycles two years, 117 (Arm-1) were completed 83 patients transferring that only morphologically assessed, 90 (Arm-2) 67 euploid biopsy-based PGT-A. Participants/materials, setting, methods Embryos cultured in 25 μL continuous until blastocyst stage. “SEM+BF” samples ongoingly collected archived at end culture. In Arm-1, after outcomes determined, corresponding pulled out niPGT-A. Arm-2, also performed some cases did up SI/LB. NiPGT-A NICS incorporated ChromGo platform (Yikon Genomics). Main results role chance Out 110 (94%) yielded informative result. Among these, 46 transferred determined “euploid” 15 resulted SI/LB; 32 “aneuploid” 27 Therefore, positive values (PPV NPV) 32.6% (15/46) 84.4% (27/32), respectively. Another Arm-1 “suspected mosaic” they showed similar rate SI/LB per those ones (37.5% vs. 32.6%, P = 0. 8092). Compared overall, “transferrable” (“euploid” + mosaic”) much higher (34.6% 27.6%, 0.3399). 39 “Euploid” PGT-A significantly better than (43.3% 0.0243). 51 SI/LB, tested 6 being 3 “abnormal/aneuploid”, raising concern possible false Limitations, reasons caution enrolled non-selected patient population inclusion few history repeated failure. Aneuploidy screening by non‐invasive prenatal testing in twin pregnancy

OBJECTIVES To describe our experience with non-invasive prenatal testing (NIPT) in twin pregnancy. METHODS Two sets of maternal blood samples from twin pregnancies were analyzed at our laboratory using NIPT: 115 stored samples from pregnancies with known outcome (Clinical Study A) and 487 prospectively collected samples for which outcomes were requested from providers (Clinical Study B). NIPT...

non-corrective approach to pronunciation

the aim of this study has been to find answers for the following questions: 1. what is the effect of immediate correction on students pronunciation errors? 2. what would be the effect of teaching the more rgular patterns of english pronunciation? 3. is there any significant difference between the two methods of dealing with pronuciation errore, i. e., correction and the teaching of the regular ...

Aneuploidy Causes Non-genetic Individuality

Phenotypic variability is a hallmark of diseases involving chromosome gains and losses, such as Down syndrome and cancer. Allelic variances have been thought to be the sole cause of this heterogeneity. Here, we systematically examine the consequences of gaining and losing single or multiple chromosomes to show that the aneuploid state causes non-genetic phenotypic variability. Yeast cell popula...

A non-invasive method for measuring preimplantation embryo physiology.

The physiology of the early embryo may be indicative of embryo vitality and therefore methods for non-invasively monitoring physiological parameters from embryos could improve preimplantation diagnoses. The self-referencing electrophysiological technique is capable of non-invasive measurement of the physiology of individual cells by monitoring the movement of ions and molecules between the cell...
